fastest is -- without outright harming yourself (for which I could be cruel and try to think up methods, but let's save you from that) -- to simply expose it often and play with it often.
I don't say it's optimal to lessen the sensitivity, and one /can/ go too far, I know I have, and I know I have less sensitivity in my glans than I should have at my age.
